The environment all looks similar in the scene where Wesker is walking down the stairs, saying "We last met...at the Spencer Estate, wasn't it?," and where he says "Poor performance indeed."
I'd have to agree that Wesker is probably speaking to Excella there. If I remember correctly, she is in her "late-20's" according to the RE5.com website. Perhaps her father was an associate of Spencer's (TriCell & Umbrella) 10+ years ago. Wesker, being a researcher, may have met Excella that way. And it would explain why Spencer is seemingly hiding out near / in the care of Excella.

Wesker probably isn't speaking Excella. She would have been a child/teen at the time of the Spencer Mansion incident. And if he was speaking to her, she would have been looking at him a bit more. Instead, she turns her head toward whoever Wesker is walking down to see. It's probably Chris and Sheva...and he's being an asshole to Chris. Of course...we also don't see the full extension of the conversation. For all we know, Wesker continues on to correct himself and mention some of the events in CV, further antagonizing Chris and starting the fight between them.

Wesker could have still met Excella back in the day. Powerful pharmaceutical mogul (Excella's father) hanging out with powerful pharmaceutical mogul (Spencer). Papa Gionne brings his intellectually-gifted daughter along for her education/enlightenment and she happens to meet some Umbrella researchers (Wesker, Birkin, et al).

You all forget that this sentence could be from a completely different scene and is only montaged that way in the trailer.
I remember, in DMC4 trailer, there was a scene where Agnus holds Kyrie and says "This is all Credo's doing" - in game he says those words long before that particular cut-scene occurs. And don't me even mention MGS trailers, where such thing is common.
