Well it could always be Ada since she seems to be hanging around a lot with Wesker. But then again, Ada is tied to Leon also. It could be Jill since I cannot believe the gravestone that Jill is dead. Carlos, Barry and Rebecca probably are since nothing has been mentioned of them since 1998 (RE timeline). As soon as someone makes a big fuss over a "death" then they probably are not dead, unless it goes the Silent Hill 2 route and we find Chris did in Jill and the Executioner (which I call Sackmid Head) and the other T-Plagas are their to punish Chris until Chris becomes so guilt ridden, he drives his car into a lake.

If Wesker is a definate show, then its probably going to be Ada.

Though it would be a real laugh if it was Claire.

Mystery Woman is almost always with Excella or "Fireworks" Guy, so either (a) she is their minion, or (b) she's on loan from Wesker as "extra muscle."

At least the Mystery Woman is throwing down while wearing a leather get-up (though a long, billowing cloak is counterproductive), as opposed to Ada fighting Spaniards, monsters, and Krauser in a slinky red dress. <-- That's just ridiculous. Please, keep Ada far away from RE5. Her Matrix-fu only belongs with NeoL.

The tube lady is also wearing a leather outfit and its dark red (See I told you I could see red!). It would be a major coincidence and poor design if both these new female characters where wearing the same outfit. I think that this is proof that the tube girl is birdlady!! Come on what do you guys think cuz I think thats pretty damn good evidence.

Realistically speaking, this is Capcom, guys. As much as we all would like to see Capcom tie this game with the stories of past games, more than likely she's another new character. Personally I hope she is Sherry. If not a whole new character, then Sherry makes the most sense.

they all appear only once in games, but we consider them like big roles?

alber and alexia were both villians in one time game they both are dead (alber at least for sure)

sadler was a one time villian that died in the same game, along with the new guys in the new rE5 game, i personally think they will die too. a point ive proven

I bet "Bird-Woman" is actually Jill in a disguise, planning on sneaking into the village in order to help Chris. The gravestone is just some sort of trick to throw players/fan's off trail so when we actually do see Jill reveal her true identity, it's an attempt to be an "OMG WTF BBQ AWSUMM!" moment. Capcom have a bigger chance at screwing things up than making things better.
