Well, in CV, the virus of the game was extracted from an arthropod. Chances are, from what we've seen in the trailers presented so far, is that the Progenitor too was extracted from an arthropod. Perhaps Alexia learned about the true origins of the Progenitor and that is what inspired her search in ancient viruses in side of Queen Ant...

I think he mentioned 2,3 and 4 because of the game play. CVX and 0 do deal with the back story of Umbrella and a lot with the virus that is going to be in 5 but CVX and 0 where harder games and aquired taste for many.
